           REAGENT PREPERATION FOR
 Zn, Fe & Cu in soil

DTPA
Take 1.967 gm AR DTPA and 1.470 gm CaCl2
               Add 25 ml Distilled Water
              
              Add 13.3 ml TEA
              
                Make up volume 1.0 Ltrs.

1.0 N Potassium Dichromate:

Dissolve 77.08 gm of Ammonium Acetate in 500 ml of D.W. and makeup Volume 1.0 Lts. Adjust the pH to 7.0 with Glacial acetic acid or Ammonium Solution.

Standard K Solutions:

Prepare 1000 mg L-1 K solution by dissolving 1.909 gm of dried (in oven) KCl salt per liter solution. Dilute volume of this a solution to get 100 ml working standards containing 10, 15, 20, 25, 30 and 40 mg KL-1. The working standards should be prepared in medium of extraction.

0.5 NaHCO2 Solution:
       
Dissolve 42 gm of Sodium Bicarbonate in 500 ml of Hot Water and Dilute to 1.0 Lts. Adjust pH to 8.5 Using Dilute NaOH or HCl

1.5 % Ammonium Molybdate Solution:

Dissolve 15 gm of AR Ammonium molybdate in 300 ml of warm water, cool and exact 350 of 10  N HCl. Make the volume to 1.0 Lts.

40 % SnCl2 Stock Solution:

10 gm stannous chloride in a 100 ml of glass beaker. Add 25 ml of Conc. HCl and dissolve by heating. Cool and store in an umber coloured bottle in dark, after adding of a small piece of Zinc Metal AR to prevent oxidation 

 From Stock Solution prepare a dilute SnCl2 Solution
(.5 ml dilute to 66 ml in D.W.) Immediately before use.

1.0 N Potassium Dichromate:
Dissolve 49.04 gm AR K2 Cr2 O7  in 500 ml D.W. Make the Volume 1.0 Lts.

  0.5 N Ferrous Ammonium Sulphate:
Dissolve 196 gm Ferrous ammonium Sulphate in D.W
Add 20 ml of Conc. Sulfuric Acid and Make the Volume 1.0 Lts.

              Diphenylamine Indicator:
              Dissolve .05 gm Dye Mixture in 20 ml in D.W.  and 100 ml Conc.
                Sulfuric Acid

        (Williams and Steinbergs, 1959 Method)

Take 10.0 gm of Soil Sample in 100 ml of Conical Flask
Add 50 ml of Calcium Chloride of 0.15 %
Shake for 30 Minutes
Filter into 100 ml of Beaker through Whatman Paper No.1
Take 20 ml of clear filtrate into a 25 ml VF
Add 2.0 ml of Glacial Acetic Acid
Add 1.0 gm of Barium Chloride and shake
                               ⬇(Shake for 2 minutes)
Add 1.0 ml of 0.25 % Gum Acacia Solution
Add 1.0 ml of 0.25 % Gum Acacia Solution
Make Volume up to 25 ml with DW and Take reading at 420 nm wavelength by Spectrophotometer

       (Lindsay and Norvell,1978 Method)


Procedure:
Take 10 gm of Soil Sample in 100 ml of Conical Flask
 ⬇
Add 20 ml Dietheline Triamine Penta Acetic Acid (DTPA) and Maintain pH 7.3 with HCL & NaOH
Shake for 2.30 Hours
Filter into 100 ml of Beaker through Whatman paper No. 41
Take Reading by Atomic Absorption Spectrophotometer (AAS) of the Filtrate

Procedure:
Take 5 gm of Soil Sample in 100 ml of Conical Flask
 
Add 25 ml of Ammonium Acetate of 1.0 Normality and Maintain pH 7.0 with Glacial Acetic Acid & Ammonium Solution
Shake for 30 Minutes
Filter into 100 ml Beaker through Whatman paper No.1
Take Reading by Flame Photometer of the Filtrate
